A Study on Estimation for Germination Activity of Fungous Conidiospores

Abstract
|
|
|
The study was designed to investigate a simple and rapid estimation, technique for germination activity of fungous conidiospores using a hanging slide culture method. The culture colonies of 4 fungal strains:
Trichophyton (T.) rubrum. T. interdigitale Priestly and Sporotrichu n beurmanni and Candida albicans were selected for study.
Each strains cultured on Sabouraud¢¥s glucose agar media were alternately observed the appearance of active growth phase of conidiospores (blastospores).
Then activated spores were picked out and inoculated in one drop of Sabouraud¢¥s glucose broth on cover glass plate, and the hanging slide culture was performed.
Following results and conclusions were made:
1. The germination rate was computed. The germination rate, of T rubrum was average 85. 3% after 24 hours of. cultivation and 93% after 48 hours. And the rate of T. interdigitale was, 89% after 24 hours and 91. 6% after 48 hours.
2. The germination rate of Candida albicans was computed at average 96.3% after 24 hours of cultivation.
3. The germination rate of Sporotrichum beurmanni was computed at average 25% after 48 hours and 26% after 72 hours of cultivation.
4. This method for study could be applied reasonably to determination of minimal inhibit concentration of anti fungul remedies.
